Blazing Saddles is a contender, for sure. I haven't seen Friday in over 20 years, so I can't judge. Monty Python movies tend to be a little uneven, so I don't think they're real contenders. Never seen Duck Soup or Stripes and I don't really care for Caddy Shack.
I think we could add Airplane and The Naked Gun to the conversation.

I think Ruddo's picked most of the objectively best. My personal Mount Rushmore would be:
Anchorman Best In Show The Big Lebowski Borat Bridesmaids Dr. Strangelove Happy Gilmore Monty Python and The Holy Grail Office Space Step Brothers There's Something About Mary Zoolander
There's also a bunch of movies that are comedic dramas, and I'd pick as being better movies, but they're not necessarily funnier. Stuff like Rushmore, Groundhog Day, Big, Friday, Dazed & Confused, Napolean Dynamite, The Truman Show, Beverly Hills Cop, Crocodile Dundee, etc

Gun to head, if I could only watch one comedy for the rest of my life, I'd have a tough time deciding between Step Brothers and The Big Lebowski.
